Lecture 1:
 
A new, fully automated on-line derivatization and cleanup solution with LC/MS-determination of Glyphosate and AMPA

Abstract:
There is widespread interest in the reliable and sensitive determination of Glyphosate residues in food and in water. The most common approach is to perform direct large volume injection of the sample, but this approach results in severe matrix influence, making the determination in food next to impossible. Other approaches rely on derivatization techniques, for water samples both pre- and post column derivatization methods are used successfully. Due to the high matrix load of most food samples, a cleanup step, for example, using solid phase extraction has to be implemented into the method.
 
In this work, a new method is presented, combining automated derivatization using FMOC-Cl (Fluorenylmethyloxycarbonyl chloride) with cleanup in a high pressure SPE system. The sample preparation procedure takes only 20 minutes and performed during the HPLC run time for the preceding sample, resulting in an overall analysis cycle of 20 min per sample once the first sample has been prepared. The derivatization process is selected step by step in the analysis software and set up as part of the overall analysis. Due to the high degree of automation every step is performed in a highly uniform manner and the variation coefficients obtained for the analysis is generally lower than for analysis methods relying on manual sample preparation procedures. Results for water samples as well as for different matrices like linseed, wheat and tea are presented with limits of determination well below 0.01 mg/kg for both glyphosate and AMPA

Automated sample preparation, matrix elimination, and analyte concentration for sensitive and accurate determination of pesticides in difficult matrices

Abstract:
An overview is given in this presentation of various automated sample preparation techniques including three different SPE techniques based on standard dimension cartridges with a packed adsorbent bed; dispersive SPE cartridges with loosely bound sorbent in disposable pipette tips; and on-line SPE based on exchangeable cartridges respectively. Additional examples are given here showing how automated SPE can be combined with analyte concentration, analyte derivatization, or addition of liquids such as internal standards to further improve limits of quantitation, selectivity, stability and quality of the analysis for both GC/MS and LC/MS analysis under integrated software control.
